Terminal sialylation of commercial rVWF concentrate is responsible for attenuated macrophage-mediated clearance via the MGL receptor

The half-life of recombinant human VWF (VWF) expressed in CHO cells has proven to be longer than plasma derived (pd)-VWF concentrates. Ciara Byrne, Ph.D., discusses the hypothesis of differences in terminal sialylation mediate the attenuated clearance of rVWF. This presentation discusses a comparison of the clearance of r- and pd-VWF in the presence or absence of clodronate-induced macrophage depletion. In conclusion, the study findings demonstrate that altered sialylation of rVWF is responsible for attenuated macrophage-mediated clearance, which may explain the extended half-life of rVWF.
